چکیده

Soybean is a rich source of protein and oil for humans. It forms symbiotic association with rhizobia to fix atmospheric nitrogen its growth development. Rhizobial type-III effectors act as important signaling molecules in symbiosis. However, they are poorly studied. In this study, mutation TtsI Sinorhizobium fredii HH103 reduced nodule numbers dry weights SN14 (cultivated soybean) ZYD00006 (wild soybean). Moreover, it nitrogenase activity but not ZYD00006. Based on the differences between inoculated mutant, two conditional quantitative trait loci associated were identified chromosome segment substitution lines derived from Using transcriptome analysis, single-nucleotide polymorphism (SNP) qRT-PCR, Glyma.13G150000 (GmTNRP1) was candidate gene soybean that activity, along NopT (a effector). addition, GmTNRP1 (an LRR-RK family located cell membrane) observed be induced by negatively regulate activity. transgenic hairy roots silencing, no significant difference terms nodules mutant those type). decreased after overexpression GmTNRP1. SNP promoter affected expression response NopT. Haplotype analysis revealed belonged Hap1 Hap2, which explained different results varieties. To best our knowledge, first study identify rhizobial effector nodules. can This enhanced understanding mechanism underlying rhizobia.

Calorimetry of nitrogenase-mediated reductions in detached soybean nodules.

Heat evolved by isolated soybean (Glycine max cv Clark) nodules was measured to estimate more directly the metabolic cost associated with the symbiotic N(2) fixation system. A calorimeter constructed by modifying standard laboratory equipment allowed measurement on 1 gram of detached nodules under a controlled gas stream. Simultaneous gas balance and heat output determinations were made. There w...

Mechanism of Nitrogenase Inhibition in Soybean Nodules : Pulse-Modulated Spectroscopy Indicates that Nitrogenase Activity Is Limited by O(2).

A novel, pulse-modulated spectroscopic system for measuring fractional leghemoglobin oxygenation and infected cell O(2) concentration (O(i)) in intact attached nodules of soybean (Glycine max) is described. The system is noninvasive and uses a pulsed (1000 Hertz) light-emitting diode coupled to an optical fiber to illuminate the nodule with light at 660 nanometer. A second optical fiber receive...

Rhizobial infection is associated with the development of peripheral vasculature in nodules of Medicago truncatula.

Nodulation in legumes involves the coordination of epidermal infection by rhizobia with cell divisions in the underlying cortex. During nodulation, rhizobia are entrapped within curled root hairs to form an infection pocket. Transcellular tubes called infection threads then develop from the pocket and become colonized by rhizobia. The infection thread grows toward the developing nodule primordi...

Properties of the nitrogenase system in cell-free extracts of bacteroids from soybean root nodules.

The capacity of excised nodules from soybean plants to fix atmospheric N2 was demonstrated convincingly in 19521 by use of the sensitive '5N technique. Although the specific inhibitory effect of H2 on N2 fixation by nodulated red clover was discovered thirty years ago,2' 3 an understanding of the biochemistry of N2 fixation in symbionts has been delayed as a result of difficulties in obtaining ...
